摘要:目的 阿司匹林对巨核细胞增殖、膜抗原表达及凋亡的影响。方法 将骨髓单个核细胞接种于含不同浓度阿司匹林的H2000培养基中,其中含Tpo 50ng·ml-1、SCF 50 ng·ml-1和IL-3 10ng·ml-1,5%CO2、370C、饱和湿度的孵育箱中培养14d。结果 高浓度阿司匹林使巨核细胞的增殖变化明显高于低浓度,巨核细胞集落(CFU-MK)数减少,细胞丛(FFU-MK)
数增加,CD41膜抗原表达率显著降低(P∠0.05);AnnexinV表达率明显增高(P∠0.05);DNA倍体分析的凋亡指数明显升高(P∠0.05);电镜观察可见出芽、空泡样结构和胞浆内颗粒减少。结论 高浓度阿司匹林可影响巨核细胞的正常代谢和促进其凋亡。

Effects of Aspirin on Megakaryocytes Multiplication in Human Bone Marrow

ABSTRACT:Aim To investigate the effects of aspirin on megakaryocytes multiplication in human bone marrow. Methods Separated megakaryocytes were cultured in vitro with medium H2000 containing aspirin 0,1.6,3.2,6.4,12.8 and 32.0mg·L-1,Tpo 50ng·ml-1,SCF 50ng·ml-1 and IL-3 10ng·ml-1 at 5% CO2,370C and saturation humidity. Using flow cytometry and observed by transmission electron microscopy to understand if the aspirin could inducing changes of megakaryocytes multiplication. Results There are more significant changes of megakaryocytes multiplication at higher concentrations aspirin than lower. The changes are reducing colony-forming unit but increasing flora-forming unit megakaryocytes,significant reducing(P∠0.05)membrane antigen CD41 expression but increasing(P∠0.05)AnnexinV and significant increasing(P∠0.05)apoptosis rate of DNA diploid analysis.The morphologic change of apoptosis cell such as budding,vacuolization and increasing granule in cytochylema were determined by electron microscopy. Conclusion Higher concentrations aspirin may interfere with the normal metabolism and accelerate apoptosis of megakaryocytes in human bone marrow.
