摘 要 目的: 探讨缬沙坦对肾性高血压大鼠血管内皮细胞的保护作用。方法: 包埋左肾制备高血压大鼠模型,给予缬沙坦治疗5周,尾容积法测动物血压,观察缬沙坦组与高血压组血压变化并应用多导电生理记录仪描记心电图,放免法测定各组血浆血管紧张素(angiot-ensin ,AngⅡ)和内皮素(endothelin, ET-1)水平,硝酸还原酶法测定一氧化氮(nitrico-xide,NO)水平。结果: 缬沙坦可使肾性高血压大鼠血压回降84%,血浆血管紧张素Ⅱ含量升高26.2%,血浆一氧化氮含量升高64.4%,内皮素含量下降49.1%(P<0.01)。缬沙坦组与高血压组心率变异各指标无明显差异(P >0.05)。结论: 缬沙坦通过促进内皮细胞分泌NO,抑制内皮细胞分泌内皮素,对肾性高血压大鼠血管内皮细胞功能具有保护作用。

Protective effects of valsartan on endothelion in rats with renal hypertension

ABSTRACT Objective: To explore the protective effect of valsartan on endothel- ion in rats with renal hypertension . Method: Make the rats model with renal hypertension , the rats were treated with valsartan for 5 weeks, Record the blood pressure and ECG. The plasma levels of NO,AngⅡ and ET were measured in each group . Result: The level of plasma AngⅡand ET with hypertension were significantly higher(P <0.01),but the plasma NO level was lower than those of controls(P<0.01).In rats with hypertension, the SBP were significantly lowered(P<0.01), the plasma level of AngⅡand NO increased(P<0.01) after the treatment of valsartan ,and the plasma level of ET decreased. Conclusion: The treatment of valsartan plays an important role in protecting the endothelial function through inhibiting endothelin excreting ET and promoting endothelin excreting NO.
