用高效凝胶色谱法测定红毛五加多糖分子量

摘要 目的:用高效凝胶色谱法测定红毛五加多糖分子量。方法色谱柱为OHPaK SB-805HQ(8mm*300mm),以高纯水为流动相,流速0.8ml.min-1,柱温35度,示差折光器检测。结果AGP1 分子量为91 000AGP2分子量为85 000AGP3分子量为45 000结论:红毛五加多糖分子量在91 000~45 000之间,AGP1AGP2AGP3三种组分组成。

Determination of  Molecule Weight of Polysaccharrides of Acanthopanax giraldii Harms by HPGPC

Abstract Objective: To determine the Molecule weight of Polysaccharride of Acanthopanax giraldii Harms by HPGPC. Methods: 1)The Molecule weights of Polysaccharrides of Acanthopanax giraldii Harms were determined by using HPGPC.2The components of hydrolysates of Polysaccharrides of Acanthopanax giraldii Harms was analyzed by using TLC. Result : The Molecule weights  of  AGP1 ,AGP2,AGP3 were 91000 85000 and 45000 , respectively . The components of hydrolysates of  AGP1 ,AGP2,AGP3 were  D-galacturonicacid,galactose,arabinose and an unknown monosaccharide; D-galacturonicacid , galactose , arabinose , xylose,rhamnose and an unknown monosaccharide ; galactose,arabinose , xylose, rhamnose and an unknown monosaccharide, respectively . Conclusion: The Molecule weights of Polysaccharrides of Acanthopanax giraldii Harms was between 91000 and 45000, and it composed by AGP1,AGP2,AGP3.
