法测定血清中更昔洛韦浓度

    目的: 建立测定血清中更昔洛韦的RP-HPLC法。方法:以阿昔洛韦为内标,采用C18柱及保护柱,流动相为甲醇-水(6040v/v),磷酸调PH6.5。流速1.0ml.min-1。紫外检测波长254nm结果:更昔洛韦与内标分离良好,更昔洛韦保留时间9.25min,阿昔洛韦保留时间为11.23min。回归方程为A1.079×102C0.202 (r=0.9997)。其线性范围为0.0510µg.ml-1结论:该法操作简便,测定结果精密度高,重现性理想,适用于药动学研究。

Reversed-Phase High-performance Liqiud Chromatographic Assay of Ganciclovir in Human Serum

ABSTRACT Objective: To set up reversed-phase high performance liquid chromatographic assay to determine the content of Ganciclovir in human serum. Method: Aciclovir was used as internal standard. Analysis was carried out on a chromatographic system using C18 column and UV detection at 254 nm. Methanol-water (60:40) was used as mobile phase, adjusted pH6.5 with phosphoric acid. Velocity of flow was 1.0 ml.min-1. Result: The Ganciclovir and internal standard departed well. The retention time of Ganciclovir and aciclovir were 9.25 min and 11.23 min. The regress equation was as follows: A107.9C0.202 (r=0.999 7). Calibration was linear from 0.05 to 10 µg.ml-1. Conclusion: This assay is easy to operate and to be repeated. It appears to be suitable for the pharmacokinetic studies.
