  肿瘤是严重威胁人类健康的常见病和多发病.目前肿瘤的治疗仍然是以化疗、放疗、手术治疗为主,其中化疗仍然是治疗肿瘤的主要手段之一,但是在肿瘤的药物化疗过程中,常能见到相同病症的不同病人对同一种抗肿瘤药物出现不同的反应,有的甚至血药浓度相同,但是疗效大相径庭.近年来,随着分子生物学、分子遗传学与分子药理学的发展,人们逐渐认识到,不同个体对同一抗肿瘤药物的不同反应,很多是源于抗肿瘤药物相关代谢酶的活性在个体间的差异,如:细胞色素P450酶3A4(CYP3A4)活性在个体间的差异可达30倍;硫嘌呤甲基转移酶(TPMT)活性在个体间的差异大于30倍;尿苷二磷酸-葡萄糖醛酸转移酶(UGT)活性在个体间的差异可达50倍.因此,在临床肿瘤化疗过程中,要重视抗肿瘤药物相关代谢酶的个体差异,在有条件的医院可以开展酶表型与基因型的测定工作,并且注意其它合用药物对酶的诱导与抑制.这样才能为抗肿瘤药物的合理应用提供强有力的保证.……
