摘 要 目的:建立同时测定丙泊酚和硫喷妥钠含量的高效液相色谱方法,观察丙泊酚与硫喷妥钠混合液中两成分含量的稳定性。方法:采用Lichrosorb C18色谱柱(200 mm×4.6 mm, 5 μm),甲醇-水(70∶30)为流动相,流速1.0 ml.min-1;检测波长222 nm,以麝香草酚为内标。将1%丙泊酚与2.5%硫喷妥钠1∶1混合液及1%丙泊酚注射液、2.5%硫喷妥钠注射液分别置冰箱4 ℃,室温25 ℃和恒温箱40℃环境中。在第0,1,3,7天分别取样,测定丙泊酚和硫喷妥钠的含量。结果:丙泊酚和硫喷妥钠的线性范围分别为10~60 μg.ml-1(r=0.999 7)和25~125 μg.ml-1(r=0.999 6),平均回收率分别为99.8%和100.7%,RSD分别为 2.4%和1.3%。1%丙泊酚与 2.5%硫喷妥钠1∶1混合液在 4 ℃和25 ℃条件下放置1,3,7 d,丙泊酚和硫喷妥钠含量下降均小于10%,在40 ℃条件下放置3,7 d,硫喷妥钠含量下降明显。结论:同时测定丙泊酚和硫喷妥钠含量的方法简便,准确。1%丙泊酚与2.5%硫喷妥钠1∶1混合液在4 ℃和25 ℃条件下稳定。

Determination of Propofol and Thiopental in a 1∶1 Volume Mixture by HPLC and Its Chemical Stability

ABSTRACT Objective: To establish a method for the simultaneous determination of propofol and thiopental by HPLC, and study the stability of propofol and thiopental mixture.Method: The Lichrosorb C18 column (200 mm×4.6 mm, 5 μm)was used. The mobile phase was methanolwater (70∶30).The flow rate was 1.0ml.min-1.The detection wavelength was at 222 nm. The internal standard was thymol. The 1% propofol and 2.5% thiopental in a 1∶1 volume mixture was maintained at 4 ℃,25 ℃ and 40 ℃, respectively. The propofol and thiopental concentrations of samples were measured at time of 0 ,1, 3, and 7 days.Result: The linear rang of propofol and thiopental was 10~60 μg.ml-1(r=0.999 7) and 25~125 μg.ml-1(r=0.999 6), respectively. The average recovery were 99.8%and 100.7% with RSD2.4%and 1.3%,respectively.The propofol and thiopental concentrations of samples have no significant changes at 4 ℃, 25 ℃ in 7 days.Conclusion: The method appeared to be simple, accurate.The 1% propofol and 2.5%thiopental in a 1∶1 volume mixture at 4 ℃, 25 ℃ was stable.
