摘 要 目的:建立氧氟沙星氢化可的松滴耳液含量测定的方法。方法:采用RP-HPLC法测定滴耳液中氧氟沙星与氢化可的松的含量。色谱柱为Poresphere C18(150 mm×4.6 mm,5 μm);流动相为甲醇-0.025 mol.L-1磷酸溶液(用三乙胺调pH至3.0)(55∶45); 流速0.8 ml.min-1; 检测波长为240 nm; 检测灵敏度为0.16AUFS,进样量20 μl。结果:氧氟沙星在30~480 μg.ml-1浓度范围内,峰面积与浓度的线性关系良好(r=0.999 9),平均回收率为99.8%; 氢化可的松在5~80 μg.ml-1浓度范围内,峰面积与浓度的线性关系良好(r=0.999 9),平均回收率为 99.9%。结论:本方法快速、简便,精密度好,重复性好,可作为氧氟沙星氢化可的松滴耳液的含量测定方法。

Determination the Content of Ofloxacin Hydrocortisone Ear Drops by RP-HPLC

ABSTRACT Objective: To determine the content of ofloxacin hydrocortisone ear drops.Method: RP-HPLC method was adopted to determine ofloxacin and hydrocortisone simultaneously.The Poresphere C18column (150 mm×4.6 mm,5 μm)was used, mobile phase consisted of methanol0.025 mol.L-1 phosphic acid solution(adjusted to pH3.0with triethylamine)(55∶45), with flow rate of 0.8 ml.min-1 and detection wavelength was at 240 nm. The detection sensitivity was 0.16AUFS and the injected volume was 20 μl. Result: The calibration curve of ofloxacin was linear within the range of 30 to 480 μg.ml-1(r=0.999 9) and the average recovery was 99.8%.The calibration curve of hydrocortisone was linear within the range of 5 to 80 μg.ml-1(r=0.999 9) and the average recovery was 99.9%. Conclusion: The method is simple, rapid, accurate and reproducible, and can be used for quality control of the preparation.
