      摘 要 目的:考察姜蚬护肝汤对酒精性肝损伤小鼠的保护作用及其机制。方法: 小鼠按体重随机为正常组,模型组,姜蚬护肝汤低、中、高剂量组,每组10只,连续灌胃给药30 d后,灌胃50%乙醇溶液14 ml·kg-1,禁食16h后取血,取肝脏;测定血清中的TNF-α、AST、ALT的水平;测定肝组织中MDA、GSH、TG、Caspase-3、Caspase-8和NF-κB的水平;HE染色后观察肝组织病理学改变。结果: 姜蚬护肝汤高剂量组小鼠的TNF-α、AST、ALT、MDA、TG、Caspase-3、Caspase-8、NF-κB均较模型组显著降低,GSH则显著提高(P<0.05);肝脏脂肪变性及组织坏死情况有所减轻。结论:姜蚬护肝汤对实验性酒精性肝损伤具有良好的对抗作用,其可能作用机制为降低TNF-α和NF-κB的过度表达从而减轻肝细胞的炎症反应,并减少Caspase-3、Caspase-8诱导的肝细胞凋亡。
英文摘要:
      ABSTRACT Objective: To investigate the protective effect and mechanism of JXHG Decoction on alcoholic liver injury. Methods: Each group of mice were fed for 30 days. Then 50% alcohol used to establish hepatic injury in mice by intragastric administration. 16 hours later, the levels of TNF-α, AST, ALT in serum and MDA, GSH, TG, Caspase-3, Caspase 8 in liver were measured accordingly. Results:JXHG Decoction could reduce the levels of TNF-α, AST, ALT, MDA, TG, Caspase-3, Caspase 8, NF-κB and increase GSH, while alleviated tissue necrosis and aelipose degeration of hepar. Conclusion:JXHG Decoction has favorable anti effects on alcoholic liver injury. Its possible mechanisms are reducing the inflammatory reaction by depressing the expression of TNF alpha and NF kappa B in liver cells, and alleviating the hepatic cell apoptosis induced by Caspase 8 and Caspase 3.
